Tadalafil was first discovered and developed by Galxo Wellcome while under the partnership of Biotechnology Company ICOS and then it was developed again and marketed as Cialis by Eli Lilly and Company. The two companies became a joint venture called Lilly ICOS, LLC. In December of 2003 the FDA approved tadalafil for sale in the US for the treatment of erectile dysfunction.
When Tadalafil is used, the man’s central nervous system stimulates the release of chemicals including nitric oxide which is the main contributor for getting and sustaining an erection. Nitric oxide stimulates production of cyclic GMP which is a chemical that relaxes the smooth muscles in the penis. This in turn allows the blood flow into the pool-like cavernous sinuses, flooding the penis.
This increased blood flow doubles the diameter of the spongy chambers and the vein surrounding the chambers are squeezed almost completely shut by the pressure. The veins are then unable to drain the blood out of the penis so it becomes erect.
After ejaculation, the cyclic GMP is broken down by an enzyme called phosphodiesterase-5 or PDE5 and other compounds are then released to make the penis become flaccid, or back to its original state.
